Plasmodium vivax infection compromises reticulocyte stability

Abstract The structural integrity of the host red blood cell (RBC) is crucial for propagation Plasmodium spp . during disease-causing stage malaria infection. To assess stability vivax -infected reticulocytes, we developed a flow cytometry-based assay to measure osmotic within characteristically heterogeneous reticulocyte and P. samples. We find that erythroid decreases erythropoiesis maturation. Of enucleated RBCs, young reticulocytes which are preferentially infected by , most osmotically stable. infection however levels close those RBC disorders cause hemolytic anemia, significantly greater degree than falciparum destabilizes normocytes. Finally, new permeability pathways contribute decreased infected-reticulocytes. These results reveal vulnerability could be manipulated allow in vitro culture develop novel therapeutics.
